#1: First and foremost, make Illustrator faster than the previous versions. You'll make everyone happy even before you add functionality.
#2: Add the ability to modify the placement of a stroke on the actual edge of an object: centered, inside or outside.
#3: Beware how much functionality you add to the program. The sheer number of features is very likely confusing most of your first time buyers. Consider making the program more modular.
